In vitro drug permeation profiles across hairless rat skin were studied by Franz diffusion cells. Skin irritation, sensitisation, and metabolic effects were investigated. In silico models were constructed to provide insight into the potential of surface-coated MPs for delivery of drugs to the skin. Particle size, zeta potential, and encapsulation efficiency were all within the appropriate ranges. The in vitro skin permeation profiles showed significant enhancement for both model drugs. The drug release profiles showed that both drugs were released over a prolonged period. The in vitro permeation profiles with and without skin were significant. The in silico studies showed the suitability of the surface-coated MPs for delivery of drugs to the skin. The results indicated a minimal effect on the metabolic activity and the Langerhans' cell, suggesting the absence of acute and chronic toxic effects in the system developed.
